1、河北医药2017年4月第39卷第8期HebeiMedicalJournal,2017,Vol39AprNo.81141doi:10.3969/j.issn.1002-7386.2017.08.005·论著·龙胆水提物对急性胰腺炎大鼠肝损伤的保护作用及机制研究吴军尚瑞【摘要】目的探讨龙胆水提物对急性胰腺炎(AP)大鼠肝损伤的保护作用及机制。方法SD大鼠40只随机分为正常组(n=8)与模型组(n=32),模型组采用经十二指肠乳头逆行胆胰管注射5%牛黄胆酸钠方法制备AP大鼠模型,造模后大鼠分为4个亚组:模型对照组(n=8)、龙胆水提物12mg/kg
2、组(n=8)、24mg/kg组(n=8)、48mg/kg组(n=8),龙胆水提物低、中、高剂量组每天分别灌胃给予龙胆水提物12mg/kg,24mg/kg及48mg/kg,正常组和模型对照组给予等体积的0.9%氯化钠溶液,连续给药1周,1次/d。给药结束后颈总动脉取血,分离血清,检测血清ALT、AST、AMY、IL-6、TNF-α水平,大鼠脱颈椎处死后剪取肝右叶1/2,于4%中性甲醛固定,HE染色观察肝脏结构变化,westernblot检测肝脏组织NF-κβ及I-κB表达水平。结果与模型对照组比较,龙胆水提物24mg/kg及48mg/kg组血清
3、ALT、AST、AMY水平及IL-6及TNF-α表达水平显著降低(P<0.01);肝组织NF-κβ蛋白表达水平显著降低(P<0.01),I-κB蛋白表达水平显著升高(P<0.01)。结论龙胆水提物能够显著改善AP导致的肝损伤。【关键词】龙胆水提物;急性胰腺炎;肝损伤;NF-κβ;I-κB【中图分类号】R576.1【文献标识码】A【文章编号】1002-7386(2017)08-1141-04Protectiveeffectandactionmechanismofwaterextractofgentianonliverdamageofratswi
