1、枸杞多糖对实验性肝癌小鼠肿瘤细胞FasL表达的影响及其抗肿瘤作用机制作者:何彦丽,杜标炎,王慧锋,苏宁,肖霞【摘要】 【目的】观察枸杞多糖(LBP)对实验性肝癌小鼠肿瘤细胞死亡分子Fas配体(FasL)表达的影响,探讨其抗肿瘤作用机制。【方法】选用昆明种小鼠,随机分为模型组,LBP低、高剂量组(剂量分别为0625、1250g·kg-1·d-1),各组均采用右侧腋下接种H22肝癌腹水型细胞复制荷瘤模型,从第3天开始按设计剂量给药,连续14d。采用苏木素—伊红(HE)染色观察各组肿瘤细胞密度、细胞核分裂计数及间质淋巴细胞浸润情况,采用免疫组化法观察各组小鼠肿瘤细
2、胞FasL表达情况。【结果】LBP低、高剂量组均可显著降低肿瘤细胞核分裂计数(P<005);LBP高剂量组可升高肿瘤间质淋巴细胞浸润程度(P<005),HE染色显示在肿瘤边缘组织内见多量淋巴细胞浸润。LBP低、高剂量组均可显著降低小鼠肿瘤细胞FasL阳性表达指数(P<005);免疫组化染色显示模型组FasL表达强阳性,LBP低剂量组部分肝癌细胞内FasL表达阳性,FasL仅在LBP高剂量组少量肝癌细胞中有表达。【结论】LBP抗实验性肝癌的效应可能与其能抑制FasL表达,减少免疫活性细胞凋亡有关。【关键词】枸杞多糖/药理学肝肿瘤/中药疗法肿瘤组织/病理学基因表

5、e.FasLexpressionwasobservedinthethreegroupswithimmunohistochemicalmethod.ResultsTumorcellmitoticcountwasdecreasedinthetwoLBPgroups(P<005),thescoreoflymphocyteinfiltrationintumorinterstitialtissuewasincreased,andlymphocyteinfiltrationwasobviousafterHEstaininginhighdoseLBPgroup(P&l
6、t;005).TheindexofFasLexpressionpositivewasdecreasedinbothLBPgroups(P<005).FasLexpressionwasstrongpositiveinthemodelgroup,andpositiveinpartiallivertumorcellsoflowdoseLBPgroupandinlesslivertumorcellsofhighdoseLBPgroup.ConclusionThe11antitumormechanismofLBPisprobablyrelatedwithth
