1、PekingUnionMedicalCollegeChineseAcademyod‘MedicalSciences摘要葛根素主要具有保护动脉血管内皮细胞,阻止动脉粥样硬化,防止血栓形成和促进心肌血液循环,减低心肌耗氧量及心脏负荷,抗心律失常,降血压,扩张冠状动脉,保护心肌超微结构,减少心肌梗死范围等作用。广泛的应用在预防及治疗心血管方面的疾病中。近年来的研究表明,炎症反应在很多心血管疾病的发生发展与持续过程中都产生了很大作用,特别是在动脉粥样硬化的发生发展过程中。本实验以二甲苯所致小鼠耳廓肿胀与大鼠棉球肉芽肿为模型,研究葛根素一般抗炎作用。建立小鼠股动脉套管导致的血管损
2、伤模型,通过Real.timePCR检Ncox.2、TNF.a、ICAM.1、MCP.1的mRNA表达,Westernblot方法检测受损血管COX.2、TNF.a蛋白表达的变化,通过组织形态学方法观察受损血管内膜面积变化。结果显示葛根素能显著抑制二甲苯所致小鼠耳廓肿胀,能显著抑制肉芽肿组织的重量,且呈量效关系。对于股动脉套管致血管损伤模型,葛根素能抑制COX.2、TNF.a、ICAM.1的mRNA表达。葛根素可抑制股动脉套管导致的COX-2、TNF.Ⅱ蛋白水平的升高。高剂量葛根素使内膜/中膜面积比显著降低,抑制内膜增生。关键词:葛根素,炎症,血管损伤,COX.2,TN
