1、小檗碱抑制兔胆固醇结石形成的作用和机制作者:王江华 陈先祥 蔡庆和 程彩涛 何志军 吴黎明【摘要】 目的观察小檗碱对兔胆囊胆固醇结石形成的预防作用和机制。方法动物模型:日本大耳白兔30只,随机等分三组:每组10只,对照组喂普通饮食;结石组,喂含1.2%高胆固醇饲料100g;实验组,同时喂饲含1.2%高胆固醇饲料100g和每日10mg/kg的小檗碱灌胃。8周后利用全自动生化分析仪酶法测定兔胆汁中的总胆固醇、磷脂、胆盐,通过Carey表计算胆固醇饱和指数(cholesterolsaturationindex,CSI),观察各组胆囊胆汁结晶情况及环氧合酶-2(cyclooxygen
2、ase2,COX2)含量。结果实验组比成石组形成胆固醇结晶机会明显减少(8/10、3/10,?字2=5.051,P<0.05);各组胆囊胆汁中CSI(对照组0.82±0.23、结石组1.64±0.27及实验组1.13±0.29);结石组COX2表达与空白组相比明显升高(47.66±1.54比28.39±1.46,P<0.05),实验组较之结石组COX2表达量有降低(37.42±2.45比47.66±1.54,P<0.05)。结论小檗碱能够抑制胆固醇结石形成和发展,其机制与调节脂质代谢,改善致石性胆汁及抑制COX2的活性等作用有关。【关键词】胆固醇结石;盐酸小檗
