1、湖南师范大学硕士学位论文不同强度运动对大鼠骨骼肌肌纤维类型的影响姓名:满维祥申请学位级别:硕士专业:运动人体科学指导教师:汤长发20100501摘要目的:检测不同强度运动后成年SD大鼠比目鱼肌和胫骨前肌中不同肌纤维类型以及MHC亚型和大鼠体重的变化情况,旨在探讨不同强度运动能否引起快、慢肌纤维之间和MHC亚型之间相互转化。方法:健康SD大鼠40只,3月龄;体重为200.2409;分成4组:安静组(C,n=10)、小强度运动组(LE,n=10)、中等强度运动组(ME,n=10)和大强度运动组(HE,n=10)。采用孙晓娟的运动方案
2、,在坡度为00的跑台上进行训练(小强度运动组:19m/minx60min,中等强度运动组:26m/minx44min,大强度运动组34m/minx34min),共八周,每次训练要求各运动组总距离基本相等。对照组和运动组大鼠分别于安静状态和运动后24小时腹腔麻醉后断头处死,取大鼠右侧后肢比目鱼肌和胫骨前肌,分别用ATP酶染色法鉴定I型和II型肌纤维;用MoticMed6.013数码医学图像分析系统分析I型和II型肌纤维的目标个数、目标面积以及各肌纤维百分比;SDS.PAGE凝胶电泳技术分离MHCI、MHCIIa、MHCIIb和MH
