3、git,NorthernAreasofPakistan.Itwasfoundtobeanaerobic,gram-positive,rod-shaped,thermophilicbacteriumthatgrewonvarioussugars,carboxylicacidsandhydrocarbonsattemperaturesbetween45°Cand75°C.Complete16SrRNAgenesequenceofthemicroorganismexhibitedhomologytovariousspeciesofgenusGeobacillus.
4、Ahighesthomologyof99.8%wasfoundwithGeobacilluskaustophilus.Apartial(0.7kbp)chaperoningenesequencealsoshowedahighesthomologyof99.4%tothatofG.kaustophiluswhereasbiochemicalcharacteristicsofthemicroorganismweresimilartoGeobacillusuzenensis.Basedonbiochemicalcharacterization,16SrRNAand
5、chaperoningenesequences,weidentifiedSBS-4SasastrainofgenusGeobacillus.StrainSBS-4Sproducedseveralextracellularenzymesincludingamylase,proteaseandlipase.Thelipaseencodinggenewascloned,expressedinEscherichiacoliandthegeneproductwascharacterized.Therecombinantlipasewasoptimallyactivea
6、t60°C+2withstabilityatwidepHrange(6–12).TheenzymeactivitywasenhancedremarkablyinthepresenceofCa.TheKmandthe−1−1Vmaxforthehydrolysisofp-nitrophenylacetatewere3.8mMand2273μmolminmg,respectively.TheabilityoftherecombinantenzymetobestableatawidepHrangemakesitapotentialcandidateforusein
