1、艾塞那肽长效微球的制备工艺及体外释放研究122222*傅军霞,王代丰,周华,邹佳,周三云,蒋朝军(1.丽水市中心医院,浙江丽水323000;2.浙江圣兆药物科技股份有限公司,杭州310051)摘要:目的开发样品制备批量易于放大的艾塞那肽长效微球制备工艺,及快速评价微球体外释放性质的方法。方法以高剪切乳化结合高压均质法制备初乳,并采用复凝聚法制备微球。工艺中初乳与凝聚液混合的步骤采用易工艺放大的静态混合法。采用将体外释放温度由37℃提高至45℃来加速评价微球中药物释放速度。结果采用高压均质法可将初乳粒径控制在约200nm以下,形成的微
2、球中药物包封率大于96.8%,1h药物突释量小于0.5%。当制备微球的批量放大5倍时,所得微球的性质与小批量样品一致。体外释放研究显示,在37℃条件下,经过约17d的延滞期后,药物可持续释放近4周。在45℃加速条件下,药物释放速度是37℃下的2.5倍,且释放曲线与37℃下基本一致,可通过加速条件快速评价微球的体外释放情况。结论以静态混合器对初乳和凝聚液进行混合来制备艾塞那肽长效微球,样品制备批量易于放大,易于实现产业化。通过加速条件可快速评价微球的体外释放性质。关键词:艾塞那肽微球;乙交酯丙交酯共聚物微球;长效微球;可生物降解微球;
