2、法:收集人肺腺癌A549细胞CM,以10%A549CM和20%A549CM加入MC3T3一E1Subclone14细胞培养液,采用CCK一8(cellcountingkit一8)法检测MC3T3一E1Subclone14细胞增殖,碱性磷酸酶(alkalinephosphatase,ALP)试剂盒检测MC3T3一E1Subclone14细胞中ALP的活性,茜素红染色检测MC3T3一E1Subclone14细胞的矿化能力,实时荧光定量PCR检测MC3T3一E1Subclone14细胞中ALP、骨钙蛋白(osteocalcin,OCN)、I型胶原蛋白1(collagentypeI
3、alpha1,Colla1)及RUNX2(runt-relatedtranscriptionfactor2)mRNA的表达。结果:MC3T3一E1Subclone14细胞培养24和48h时,10%A549CM组和20%A549CM组的细胞增殖率均高于不加A549CM的对照组(P<0.05):10%A549CM组和20%A549CM组MC3T3一E1Subclone14细胞中ALP活性低于对照组(P<0.05);A549CM组矿化结节的数量和面积均小于对照组(P<0.05);A549CM组MC3T3一E1Subclone14细胞中ALP、OCN、Collal和RUNX2mRN
