2、法绘制细胞生长曲线,比色法检测细胞内还原性烟酰胺腺嘌呤二核苷酸磷酸(NADPH)、还原性谷胱甘肽(GSH)含量,荧光探针检测细胞内活性氧(ROS)含量;构建裸鼠动物模型,绘制肿瘤生长体积曲线与重量柱形图。结果表达IDH1“突变体的胶质瘤细胞生长明显抑制,免疫荧光染色以及TUNEL法证实细胞增殖减少、凋亡增多;细胞内NADPH水平降低伴随GSH含量降低和ROS蓄积,HO(1mmol/L)可明显增加胶质瘤细胞的生长抑制作用,GSH(1mmol/L)则可减弱抑制作用。结论IDH1基因R132H突变可显著
