1、中华临床师杂志f电子版)2013年7月第7卷第l4期ChinJClinicians(ElectronicEdition),July15.2013.v01.7.No.14·基础论著·多核巨细胞在小鼠黑色素瘤细胞转移中的作用机制研究米蕊芳刘福生金贵善【摘要】目的研究多核巨细胞在黑色素瘤转移中的作用及其相应分子机制。方法用聚乙二醇细胞融合法体外获取黑色素瘤多核巨细胞,通过流式分选黑色素瘤多核巨细胞。小鼠皮下及尾静脉注射黑色素瘤多核巨细胞并用游标卡尺测量皮下肿瘤体积,电子天平称重荷瘤肺组织以定量肿瘤转移能力。Affymatrix基因芯片分析差异表达的基因,IngenuityPathw
2、aysAnalysis(IPA)软件进行基因功能分析。结果细胞融合并流式分选成功获得黑色素瘤多核巨细胞。黑色素瘤多核巨细胞皮下生长速度减慢但肺转移能力明显增强。黑色素瘤多核巨细胞与转移相关的13-Tubulin组基因表达明显升高。黑色素瘤多核巨细胞的染色体呈现稳定性。结论黑色素瘤多核巨细胞促进黑色素瘤细胞转移能力,黑色素瘤多核巨细胞与转移相关的fI-Tubulin组基因表达升高。【关键词】巨细胞;黑色素瘤;肿瘤转移;小鼠Melanomamultinucleatedgiantcellsacquiredenhancedmetastasispotential^礁Rui-fang,L
