1、中华老年心脑血管痫杂志201t年1月第l6卷第l期ChinJGeriatrHeartBrainVesselDis,jan20l4,Vol16·No.1.基础研究.RockⅡ在大鼠脑老化中作用机制的研究胡金华,吴玉泉,赵园园,郑伟,成军,卢小军摘要:日的探讨Rock1对大鼠脑老化和海马组织凋亡作用的影响。方法选择3月龄Wister雄性大鼠8O只,随机分为脑老化组、对照组、脑老化限食组和对照限食组,每组2O只。1()周后进行为期7d的水迷宫实验,采用免疫印迹法检测各组RockII及BcI一2、Bax蛋白,并分析Rockli水平与大鼠逃避潜伏期的相关关系。结果与脑老化组比较,脑老化
2、限食组、对照组和对照限食组RockⅡ表达明显降低(P<().05。P<0.01),脑老化限食组Bcl一2明显升高,Bax和caspase一3明显降低,逃避潜伏期明显缩短,记忆搜台游泳路程明显延长,差异有统计学意义(P<0.05)。RockII与逃避潜伏期呈正相关(r一0.952,P<0.()1)。结论Rock1I可通过抑制Bcl一2表达及激活Bax蛋白表达而诱发细胞凋亡,大鼠脑老化程度与RocklI的表达水平呈正相关。关键词:Rho相关激酶类;海马;细胞凋亡;半胱氨酸天冬氨酸蛋白酶3;脑老化RoleofRockⅡinratbrainagingHUJin—hua,WUYu—qu
