2、B)表达的影响,探讨亚低温的脑保护作用及可能机制。方法:将45只sD大鼠随机分为假手术组(Sham组),脑损伤组(TBI组),亚低温治疗组。采用Mamamu’s法建立创伤性脑损伤大鼠模型,损伤后立即开始诱导亚低温,持续3h,于伤后1d、3d、5d分别处死各组大鼠留取脑组织。采用干湿重法测量脑组织含水量,免疫组织化学法及Westernblot法检测各组大鼠脑组织TNF.o/_、IL—lB及NKB的蛋白表达情况。结果:与Sham组相比,TBI组大鼠脑组织含水量升高(P<0.05),3d达水肿高峰,TNF—ot、IL.1B和NF—KB的表达显著增加(P<0.05),均为1d达高峰,持续高表
3、达至5d;与TBI组相比,亚低温治疗组大鼠各时间点脑组织含水量降低(P<0.05),TNF一仅、IL一113和NF-KB的表达明显减少(P<0.05)。结论:高表达的TNF一、IL一1p和NF.KB参与了TBI的病理过程。亚低温治疗可通过下调NF.KB信号分子,减轻创伤性脑损伤后脑组织的炎性反应,缓解脑水肿,进而起到脑保护作用。[关键词]亚低温;创伤性脑损伤;脑水肿;大鼠EfectofmildhypothermiaonexpressionofTNF-0【,IL-1I3;andNF-KBaftertrau—maticbraininjuryinratsWANGYa—Nan,ZHANGYo
