3、ninretinalglialcells(RGCs)inocularhypertentionrats.MethodsTheocularhypertentionmodelswereestablishedbycauterizingthelimbus2drainingveinsintherighteyesof42SDrats,andaconjunctivalincisioninthelefteyesoftheratsservedastheshamgroup.Theintraocularpressure(IOP)wasmeasuredwiththeTono2PenXLton

4、ometer.ThenumberofRGCsintheratswithocularhypertentionwascounted.TheexpressionofthenestinproteininRGCswassemi2quantitativelyanalyzedusingWesternblot,andtheinductionoftheexpressionofnestininMüllercellswasdetectedbyimmunochemistry.Doubleimmunofluorescencewascarriedouttoevaluatetheexpressi

5、onofnestin/GSornestin/GFAPinthefootplateofMüllercellsundertheconfocallaserscaningmicroscope.ResultsSignificantdifferenceswerefoundintheIOPbetweenthemodelgroupandtheshamgroupatvarioustimepoints(P<0105).In1weekto3weeksafteroperation,thenumberofRGCssignificantlydeclinedinthemodelgroupcomp

6、aredwiththeshamgroup(P<0105).Immunochemistryshowedthatfrom2hoursthrough1weekafteroperation,theexpressionofnestinwasgraduallyenhancedinthemodelgroupincomparisonwiththeshamgroup.Westernblotrevealedthattheexpressionofthenestinproteinreflectedasimilartendencytothatofimmunofluorescence.Thep

7、resentresultsdeterminedthatMüllerglialcellsreactedvigorouslytoincreasedintrocularpressureasmanifestedbytheinducedexpressionofnestin.ImmunoelectronmicroscopyalsoconfirmedtheinducedexpressionofnestininMüllerglialcellsinocularhypertention.ConclusionThepresentresultsuggeststhattheinduced
