3、237,China)[Abstract]ObjectiveToconstructangiogenesis—。specificRGD10—。NGR9dual’。targetingsuperparamagneticironoxidenan—-oparticies,andtoobservethephysicalproperties.MethodsDual—targetingpeptidesRGD10一NGR9weredesignedandsyn—thesized.Fluorescencelabeled

4、RGD10NGR9peptidesweresynthesizedbyincorporatingflouresceiniso—thiocyanate(FITC)intopeptides.Humanumbilica1veinendotheliacells(HUVEC)wereincubatedwithFITClabeledpeptidesandsubse—quentlytestedbyinvertedmicroscope.Ultrasmallsuperparamagneticironoxide(US

5、PIO)nanoparticleswithnarrowsizedistributionhavingaveragediametersof8—12nmweresynthesizedbyco—precipitationchemicalmethod.andthesurfacewasmodifiedtobehydrophilicbycoatingwithdextran.Then,dextranwasoxidizedandcrosslinkedtoUSPI()bypropyl—eneoxide,andthe

6、hybridpeptidesRGD10一NGR9werefinallyconjugatedtothesurfaceofUSPIOinSchiffbaseswithdex—tran.Themorphologyofpreparednanoparticleswasobservedunderelectronmicroscope.Themeanparticlesizeandsizedistributionwereobservedbydynamiclightscattering.Thesurfacecoat

7、ingmaterialsweredeterminedbyinfraredspec—trometer.Zetapotentialwastestedandmagneticintensitywasexaminedbyvibratingsamplemagnetometer.ResultsInvitrostudiesdemonstratedabetterbindingaffinityofFITC-RGD10一NGR9toHUVEC.RGD10一NGR9targetedUSPIOnanoparticlesw

8、eresuccessfullyconstructed.Thetransmissionelectronmicroscopyofthenanoparticleswashomogeneousinsizeandroundinshape,withaveragediametersof30nm.ThestablepropertiesweretestedwithZetapotentialof一60.9mVandmagneticintensityof53.54emu/gunder70~C.ConclusionRG
