1、·84-JHenanUnivSciTech(MedSci)June2014Vo1.32No.2Folin·Ciocalteu法测定山茱萸果核水溶性提取物多酚含量的研究杨晖,和素娜’,杜景霞,李艳,路西明,王建刚,吕茂平摘要:目的以没食子酸为标准物质,研究Folin—Ciocalteu法测定山茱萸果核水溶性提取物多酚含量的优化条件。方法以吸光度为评价指标,分别研究了检测波长、Folin—Ciocaheu试剂用量、NaCO用量、反应温度及反应时间对测定结果的影响。结果优化测定条件:检测波长760nm,0.4mL样品加入Folin—Ciocaheu试剂1.0mL、
2、10%Na2c0,溶液3.2mL,反应温度35℃,显色反应时间1.0h,吸光度与多酚含量在0—250~g/mL范围内呈良好的线性关系(Y=0.0053x+0.0389,R=0.9993)。方法的加标回收率为98.5%一103.0%,相对标准偏差为1.9%~2.3%(vt=6)。三批山茱萸果核水溶性提取物多酚含量平均值为39.24%。结论Folin.Ciocaheu法在本研究建立的优化测定条件下显色灵敏、准确可靠、稳定性高、简便易行,适用于山茱萸果核水溶性提取物多酚的含量测定。关键词:Folin—Ciocaheu法;山茱萸果核;水溶性提取物;多酚中图分类号:R
