1、超氧化物歧化酶和U74389G对新生鼠高氧肺损伤的治疗作用作者:王丽珍,钱燕,卢洪胜,翁寿向,卢洪萍,罗芳【摘要】目的:探讨抗氧化剂治疗高氧肺损伤的疗效及机制。方法:将新生SD大鼠随机分为对照组、高氧组、SOD组和U74389G组,每组14只。复制高氧肺损伤模型,用SOD和U74389G进行治疗。黄卩票吟氧化酶法测定SOD活性,硫代巴比妥酸法测定MDA含量,TUNEL法检测细胞凋亡,观察肺病理变化以及放射状肺泡计数(RAC)o结果:高氧组的肺系数、MDA含量和凋亡指数显著高于对照组,RAC明显减少(均P<0.01);U74389G
2、组及SOD组与高氧组相比,肺系数下降、MDA含量减少、SOD活性增高、凋亡指数下降,RAC升高(P<0.01,P<0.05)o高氧组肺组织严重水肿,中性粒细胞浸润,SOD组肺组织病理改变明显减轻,U74389G较高氧组有所减轻。SOD组在缓解肺系数、降低SOD活性及抑制凋亡方面优于U74389G组(P<0.01,P<O.05);U74389G组在抑制MDA含量方面优于SOD组(P<0・01),而两组间RAC差异无显著性。结论:抗氧化剂U74389G和SOD能减轻高氧肺损伤。【关键词】咼氧;肺损伤;新生鼠;超
