1、1225VALIDATIONOFCOMPENDIALPROCEDURESTestproceduresforassessmentofthequalitylevelsofpharmaceuticalarticlesaresubjecttovariousrequirements.AccordingtoSection501oftheFederalFood,Drug,andCosmeticAct,assaysandspecificationsinmonographsoftheUnitedStatesPharmacopeiaandtheNationalFormularyconstitut
2、elegalstandards.TheCurrentGoodManufacturingPracticeregulations[21CFR211.194(a)]requirethattestmethods,whichareusedforassessingcomplianceofpharmaceuticalarticleswithestablishedspecifications,mustmeetproperstandardsofaccuracyandreliability.Also,accordingtotheseregulations[21CFR211.194(a)(2)],
3、usersofanalyticalmethodsdescribedinUSP–NFarenotrequiredtovalidatetheaccuracyandreliabilityofthesemethods,butmerelyverifytheirsuitabilityunderactualconditionsofuse.RecognizingthelegalstatusofUSPandNFstandards,itisessential,therefore,thatproposalsforadoptionofneworrevisedcompendialanalyticalp
4、roceduresbesupportedbysufficientlaboratorydatatodocumenttheirvalidity.Thetextofthisinformationchapterharmonizes,totheextentpossible,withtheTripartiteInternationalConferenceonHarmonization(ICH)documentsValidationofAnalyticalProceduresandtheMethodologyextensiontext,whichareconcernedwithanalyt
5、icalproceduresincludedaspartofregistrationapplicationssubmittedwithintheEC,Japan,andtheUSA.SUBMISSIONSTOTHECOMPENDIASubmissionstothecompendiaforneworrevisedanalyticalproceduresshouldcontainsufficientinformationtoenablemembersoftheUSPCouncilofExpertsanditsExpertCommitteestoevaluatetherelativ
6、emeritofproposedprocedures.Inmostcases,evaluationsinvolveassessmentoftheclarityandcompletenessofthedescriptionoftheanalyticalprocedures,determinationoftheneedfortheprocedures,anddocumentationthattheyhavebeenappropriatelyvalidated.Informationmayvarydependinguponthetypeofmethodinvolved.Howeve
7、r,inmostcasesasubmissionwillconsistofthefollowingsections.Rationale—Thissectionshouldidentifytheneedfortheprocedureanddescribethecapabilityofthespecificprocedureproposedandwhyitispreferredoverothertypesofdeterminations.Forrevisedprocedures,acomparisonsho
