2、ease)相结合的技术构建了栽培大豆高盐胁迫下根部组织的酵母双杂交cDNA文库。提取高质量的总RNA,利用Oligo(dT)引物反转录合成cDNA后,经DSN处理,然后利用Advantage2聚合酶进行长片段PCR扩增,电泳检测显示,产物均一,无高丰度条带,片段范围在0.75~5.[)(】kb;RT—PCR结果显示,GmActin基因在均一化后表达明显降低,表明均一化效果较好;最后通过SMART同源重组交换技术在酵母菌株Y187中构建了酵母双杂交所需的cDNA文库。文库质量检测结果表明:cDNA文库的总独立克隆数为5.61X10cfu、文库滴度为3.4X10cfu/mL、
