2、ph,Ontario,CanadaAcceptedFebruary12,1990Summary.Parker'sratcoronavirus(PRC)isanaturally-occurringviralinfectionofthelaboratoryrat.Onthefirstpassage,ATCCstrain8190ofPRCreplicatedinL-2cells.UsingthetenthpassageofPRCinL-2cells,thecharacteristicsoftheviruswerecomparedwithpreviousstudiesofsialoda
3、cryoadenitisvirus(SDAV)replicatedinL-2cells.Basedonlightandimmunofluorescencemi-croscopicexaminationofcontrolandinoculatedcellcultures,PRC-associatedCPEwasfrequentlyconfinedprimarilytoindividualcells,andtherewererel-ativelyfewsyncytialgiantcells.Maximumtiterswererecoveredat36hpostinoculation
4、(pi).InfectiousviruswasdemonstratedatpHvalues:rangingfrom6.0to9.0andapHof7.5wasdeterminedtoproducethehighesttitersofPRC.Theoptimumtemperatureforviralreplicationwas33°C.Upto15passagesofPRCinL-929cellsfailedtoproducedetectablevirus.However,afterad-aptationinL-2cells(20thpassage),PRCreplicatedt
5、ohightitersinL-929cells.Previously,invitrostudiesofratcoronaviruseshavebeenhamperedbythelackofanidentifiedcontinuouscelllinetoreplicatethesevirusesinthelab-oratory.L-2cellsrepresentareadily-availablecontinuouscelllinethatcansupportthereplicationofrelativelyhightitersofPRC.IntroductionParker'
6、sratcoronavirus(PRC)andsialodacryoadenitisvirus(SDAV)aretwonaturally-occurringcoronavirusesinthelaboratoryratthathavebeenidentifiedandpartiallycharacterized[1,9].Inthecurrentliteraturetheyareassociatedwithdifferentpatternsofdisease.PRCisconsideredtoproduceadiseaseprimarilyoftherespiratorytra
7、ct[2,9-1,whileSDAVinfectionsareassociatedwithlesionsofthesalivaryandlacrimalglands[7],andrespiratorytract[13-1.However,thevirusesarecloselyrelatedantigenically[1].Thusconventionalserologicaltechniquesdonotservetodifferentiatebetweenthedifferentratc
