3、stemsdevelopedsofarhavelimitedthegrowthinknowledgeaboutRCVsbynotpermittinggenerationofplaque-clonedvirusstocks,reliableisolationofRCVsfromrattissues,orgrowthofhightiteredstocksofallisolates.Duetothefactthatlessthan20%ofL2(Percy)cellswerebecominginfecte
4、d,sublineswereproducedandselectedformaximalgrowthofRCVs.Screeningof238cellsublinesyieldedL2p.176cellswhichwerehighlysusceptibletoallRCVstested;however,susceptibilitydeclinedafter30passagesinvitro.Low-passagedL2p.176cellsWereusedtoisolatevirusfromnatura
5、loutbreaksandtopropagateindividualRCVplaquesintohightiteredstocks.ProteinsfromsixRCVisolateswereimmunoblottedusingpolyclonalratandmouseantibodiestosialodacryoadenitisvirusandpolyclonalmonospe-cificrabbitandgoatantibodiesagainstthepeplomer(S)andnucleoca
6、psid(N)proteinsofmousehepatitisvirus(MHV).Proteinsoftwoprototype,oneJapaneseandthreewildtypeRCVswereexaminedandfoundtobesimilartothoseofMHV,althoughtheexactsizesandratiosofproteinformswereuniqueformostRCVisolates.Thisstudyreportsthedevelopmentofacontin
