3、ndrelatedNordenVaccine(BCV-Vaccine)strainsweredeterminedusingthepolymerasechainreaction(PCR)toamplifycDNAsobtainedbyreversetranscriptionofviralRNA,andtoproducesinglestrandcDNAsforDNAsequencing.TheSgenesequencesoftheseviralstrainswerecomparedwiththose
4、ofrecentlypublishedstrainsBCV-Mebus,BCV-Quebec,andBCV-F15.Anopenreadingframeof4092nucleotides,encodingaproteinof1363aminoacidresidues,wasfoundinallsixstrains.FrameshiftsandinsertionsordeletionswerenotobservedexceptfortheBCV-F15.TheSgenesequenceswerem
5、orethan98%conservedoverallinspiteofdifferentoriginsofthesixviruses.Therewere45to56ntdifferencesbetweenthevirulentandavirulentgroupswhiletherewere6to14ntdifferencesamongfouravirulentstrains.ComparisonofthededucedaminoacidsequencesindicatedthattheSprot
6、einshadtypicalpropertiesofmembraneglycoproteins.NineteenN-linkedglycosylationsiteswerepredictedinfivestrains,and18ofthemwereconservedintheavirulentstrainBCV-L9.ThesequenceKRRSRRatthepredictedproteolyticcleavagesitewasidentifiedinfivestrainswhilethese
7、quenceKRRSVRwasfoundinBCV-F15.Substitutionsoffewaminoacidsintheputativefusogenicdomainsandtwoprolinesat507and567intheantigenicdomainsmaycausealteredimmunogenicandotherfunctionalpropertiesoftheSproteinsspecifiedbythevirulentandavirulentBCVstrains.Nine
