3、RCV)mRNAspeciesshowedthatmRNAs2and3weresmallerthanthecorrespondingtransmissiblegastroenteritisvirus(TGEV)mRNAspecies(Pageetal.(1991)J.Gen.Virol.72,579-587).SequenceanalysisshowedthatmRNA3wassmallerduetothepresenceofanewputativeRNA-leaderbindingsiteupstreamof
4、thePRCVORF-3gene.However,thisobservationdidnotexplainthedeletionobservedinPRCVmRNA2.Polymerasechainreaction(PCR)wasusedtogeneratecDNAfromthe3codingregionoftheputativepolymerasegenetothepoly(A)tailofPRCVforcomparisontotheequivalentregionfromTGEV.ThePRCVSprote
5、inwasfoundtoconsistof1225aminoacids,whichhad98%similaritytotheTGEVSprotein.However,thePRCVSgenecontaineda672nucleotidedeletion,corre-spondingto224aminoacids(residues21to245inTGEVSprotein),59nu-cleotidesdownstreamoftheSgeneinitiationcodon.ThePRCVgenomefromthe
6、ORF-3genetothepoly(A)tailwassequencedforcomparisontoTGEVinordertoidentifyotherpotentialdifferencesbetweenthetwoviruses.FourORFswereidentifiedthatshowed98%similaritytotheTGEVORF-4,M,NandORF-7genes.NootherdeletionsoranyPRCVspecificsequenceswereidentified.Corre
