2、YInvitrostudieswereundertakentoexaminetheeffectsofnon-immuneporcineperipheralbloodleukocytes(PBL)onacoronavirusinfectionduetotransmissiblegastroenteritisvirus(TOBV).TheassayconsistedofTOBV•infectedepithelialcellsexpressingviralantigensonthecellsurfaceandproducinglowamountsofinterfero
3、n(IFN).Non-immunePBLwerefoundtolimitvirusreplicationataneffector-to-targetratioof100/1evenwheneffectorcellsweredepletedofphagocyticcells.Neutralizinganti-IFNanti•bodiesdidnotabrogatetheeffect.PBLfromnewbornanimalswereaseffectiveasadultcells,whereasfibroepithelialcells,humanandmousely
4、mphoidcellsdidnotexertantiviraleffects.Undersimilarconditions,PBLfromadultanimalscouldlyseTOEV-infectedcellseveninthepre•senceofanti-IFNantibodies.However,newbornPBLwerenotcytotoxic.Moreover,depletionofNKcellsbymonoclonalantibodiespluscomplementdidnotaltertheinhibitoryeffect.Theselat

6、RDIEREINTRODUCTIONNon-specificdefencemechanismsagainstviralinfectionsincludeinterferon(IFN)productionandcell-mediatedreactions.Non-immunemononuclearcellshavebeenshowntocounteractvirusspreadingbyatleastthreedistinctmechanisms:IFNproduction,spontaneouslysisofvirus-infectedcells(ornatur
7、alkilling,NK[14,33])andinhibitionofvirusreplication[26].Insomecases,suchasMHV3orHSV1infections,thegreatsusceptibilityofyounganimalswascorrelatedwiththeirimmaturecell-mediatedimmunereactions[16,32].Inthepresentreport,wehavestudiedtransmissiblegastroenteritisvirus(TOBV),acoronaviruswhi
