3、V,aspike(S)genedeletionmutantofTGEVwithanalteredrespiratorytissuetropism,causesmildorsubclinicalrespiratoryinfections.ComparisonsofTGEVandPRCVstrainssuggestthattropismandpathogenicityareinfluencedbytheSgeneandORF3,respectively.Recently,outbreaksofTGEofre
4、ducedvirulencewerereportedinthefield.WeinvestigatedasimilarsuspectTGEVoutbreakofreducedvirulenceinnurserypigsfromaswineherdintheMidwest.ATGEVstrain(BW021898B)wasisolatedinswinetesticularcellsfromgutcontentsofadiarrheicpigandthreePRCVstrains(BW126,BW154,B
5、W155)wereisolatedfromnasalswabsfromnormalTGEV-seronegativesentinelpigsincontactwiththediarrheicpigs.SequenceanalysisoftheTGEVisolateinthepartialSgeneandORF3/3aandORF3-1/3brevealedhighhomologywithen-teropathogenicTGEVstrains.Gnotobioticpiginoculationandh
6、istopathologicalresultsrevealedthatthisTGEVisolateretainedvirulenceeventhoughinthefieldoutbreakthediarrhealdiseasewasofreducedseverity.SequenceanalysisoftheSgenedeletionregionofthethreePRCVisolatesrevealedidenticaldeletionsbe-tweennt105–752,whichdifferfr
7、omdeletionspreviouslyreportedamongPRCVstrains.ThethreePRCVisolateshadvariablesequencechangesinORF3/3aandORF3-1/3b,affectingtheORFsizeandaminoacidsequence.Thus,sequenceanalysisandpathogenicitystudiesindicatethatthisTGEVisolateresemblesotherenteropathogen
8、icTGEVstrains.Therefore,thereducedseverityofTGEobservedinthisherdmaybeduetotheongoingPRCVinfections,whichinduceantibodies1134L.Kimetal.cross-reactivewithTGEVandresultindecreaseddiseaseseverity.Theresultsoutlinedinthisstudyhighlig
