4、形成分子量大小不等的随机限制性酶切片段,将特定的人工合成的短的双链接头连在这些片段的两端,形成一些带接头的特异片段,通过接头序列和PCR引物3ˊ端选择性碱基的识别,对特异性片段进行预扩增和选择性扩增。结果只有那些两端序列能与选择性碱基配对的限制性酶切片段才能被扩增;最后将选择性扩增产物在高分辨率的变性聚丙烯酰胺凝胶上电泳,寻找多态性扩增片段扩增片段长度多态性(AFLP)特点:1)非常高的基因型分析效率;2)不需要标记开发成本,但是需要优化特异引物及引物组合;3)不需要基因组DNA序列信息;4)AFLP分析只需要很少量的DNA;5)AFLP可应用于任何生物AFL
