1、甘遂毒性的血浆代谢组学研究杨永霞1,唐冰雯2,丁佳佳2,王琳琳】,毋福海3,宋粉云2(510006r州,广东药学院:基础学院】,药科学院2,公共卫生学院3)[摘要]目的研究甘遂毒性致人鼠血浆代谢组的变化,探讨代谢组学在中药毒性研究中的可行性。方法选取SD大鼠18只,适应1周后随机分为3组:对照组、甘遂低剂量组(7.875g/kg)和甘遂高剂量组(15.75g/kg),持续给予甘遂2周后,取各组人鼠血浆,收集各组血浆样品的]HNMR谱,运用正交最小二乘判别(orthogonalpartialleast-squares-discriminantanalysis,O
2、PLS-DA)方法分析。结果与对照组相比,甘遂组血浆代谢有明显变化。甘遂低剂量组血浆中乳酸和肌酸的含量升高,月旨、3-疑丁酸盐、牛磺酸和葡萄糖的含量降低。此外,丙氨酸、0-乙酰糖蛋白和氧化三甲胺在高剂量甘遂组中升高,而不饱和脂肪酸则降低。结论本实验从分子水平阐明了甘遂毒性引起的血浆代谢组变化。[关键词]甘遂;代谢组学;核磁共振氢谱[中图法分类号][文献标志码]A[基金项目]国家自然科学基金(21005022);广东省自然科学基金(S2011010002512)[通信作者]杨永霞,电话:(020)39352197,E-mail:sheepma@163.com宋粉
