1、摘要B淋巴细胞刺激因子(Blymphocytestimulator,BLyS)是TNF家族中的一员,又称为B细胞激活因子(BcellactivatingfactorbelongingtotheTNFfamily,BAFF)。在体内外实验中,可溶性BLyS(sBLyS)能够特异性作用于B细胞,诱导B细胞分化、增殖和存活。如果敲除BLyS,小鼠体内的T1期B细胞相对正常,T2期和成熟B细胞缺失。因此,BLyS在B细胞的成熟过程中发挥至关重要的作用。而在BLyS过量表达时,小鼠表现出自身免疫性疾病样症状,提示BLyS
2、与自身免疫性疾病的发生密切相关。新近的研究发现,部分B细胞型肿瘤细胞能够合成BLyS。分泌的BLyS,可能与自身细胞的BLyS受体结合,形成自分泌途径,参与B细胞肿瘤的生长与存活。因此,BLyS的拮抗剂可能成为治疗自身免疫疾病和某些肿瘤的新型药物。首先,我们根据BLyS与其受体的相互作用结构,设计出五个BLyS的突变体,.以pET22b(+)./sBLyS为模板扩增BlyS的突变体,然后,将突变体基因酶切下来,连接到原核表达载体pET32a(+),进行表达、纯化,得到五个BLyS突变体蛋白(M1、M2、M3、M
3、4、M5)。我们设计了两组实验检测BLyS突变体的功能。第一组实验:通过MTT比色法,用BLyS及其突变体刺激小鼠脾细胞来检测他们的功能;第二组实验:将BC/PC质粒(BC/PC质粒为可表达BCMA蛋白的质粒)和NF—KB-luc质粒共转染进入293T细胞,当BCMA在293T细胞表面表达后,用BLyS及其突变体刺激,根据检测的荧光值的不同来判断有意义的突变位点。根据两组实验的结果,我们得到BLyS突变体M2、M4、M5的突变位点有意义,其中突变体M5的突变位点最有意义,这将为我们根据确定的突变位点设计BLyS
