2、ublishedonlineJanuary31,2013)(7)Over80yearsago,Warburgdiscoveredthatcancercellsgeneratecells.ThisreviewcoverstheaccumulatedfindingsregardingATPthroughtheglycolyticpathway,eveninthepresenceofoxy-keytranscriptionfactorsandmetabolicenzymesthatinducegen.Thefindin
3、gofthisphenomenon,termedthe“Warburgalterationsincancermetabolism,whichcanbeattractivetar-effect,”stimulatedmuchresearchontumorigenesis,butfewgetsforcancertherapy.explanationswereforthcomingtoexplaintheobservation.Recently,advanceddevelopmentsinmolecularbiol
4、ogyandhigh-TheWarburgEffectthroughputmolecularanalyseshaverevealedthatmanyofthesignalingpathwaysalteredbygenemutationsregulatecellOneoftheprominentcharacteristicsofrapidlygrowingtumormetabolismincancer.Furthermore,mutationsinisocitratedehy-cellsistheircapac
5、itytosustainhighratesofglycolysisfordrogenase1and2wereshowntoelevate2-hydroxyglutarate,ATPgeneration,irrespectiveofoxygenavailability.Thisphe-(2,8,9)whichledtochangesina-ketoglutarate-dependentdioxygenasenomenonisknownastheWarburgeffect.Thischaracter-enzyme
6、activity,resultinginanincreasedriskofmalignantisticseemstobeageneralpropertyofhighlymalignant(10)tumors.Althoughthesefindingsledtoarenewedinterestincan-tumors,independentoftheircarcinogenicorigin.Infact,cermetabolism,ourknowledgeonthespecificsoftumormetab-sig
7、nificantactivationofglycolysiswasobservedinhumanolismisstillfragmented.Thispaperreviewsrecentfindingscancertissuesthroughtheuseofaleading-edgemetabolomicrelatedtokeytranscriptionfactorsandenzymesthatplayanprofilingmethodbasedoncapillaryelectrophoresismassspec-
