2、amilymemberVI(PAQR6)isanewmemberoftheProgestinandadipoQreceptors(PAQRs)familythatfunctionsasanimportantfactorinthenongenomicactionsofrapidsteroidresponse.ConsideringtheimportantroleofPAQR6inthesenongenomicactions,wedeterminedporcinePAQR6expressionandregulation.WeclonedthecDNAofporcine
3、PAQR6andanalyzeditsgenomicstructure.SubcellularlocalizationanalysisshowedthatPAQR6-greenfluorescentproteinfusionproteinwasdistributedincytoplasm.SpatialexpressionpatternsanalysisrevealedthatporcinePAQR6wasmorehighlyexpressedinsmallintestinalthaninothertissues.Toinvestigatetheregulatory
4、mechanismofporcinePAQR6,weclonedapproximately1.5kbofporcinePAQR65’-regulatoryregionandgeneratedsequentialdeletionconstructsandevaluatedtheiractivityinadual-luciferasereporterassay.Theresultssuggestedthatthecorepromoterregionofthisgeneislocatedinthefirstexonregionfrom+90to+241.Site-dire
5、ctedmutagenesisindicatedthatadi-ponectinreceptorprotein1,E2Ftranscriptionfactor1,andSp1transcriptionfactormightbeimportanttran-scriptionfactorsforporcinePAQR6.ThisstudyisthefirstattempttoelucidatethetranscriptionalregulationofporcinePAQR6,whichestablishedafoundationforfurtherstudyandco
6、ntributedtothedeeperinvestigationoftheregulationandfunctionofPAQR6.Introductiontialfortherapidactionsofprogestin(Boonyaratanakornkitetal.,2001;Fernandes,2005).Adiponectin(alsonamedAdi-NongenomicactionsareimportantforrapidsteroidpoQ,GBP28,apM1,andAcrp30),amemberofbiologicallyresponseby
