3、endogenousregulatorynetworkisdistributedandutilizedbycellstransitioningthroughakeydevelopmentalevent.Results:Hereweinvestigatethetemporaldynamicsofasingle-celltranscriptionalnetworkof45transcriptionfactorsinTHP-1humanmyeloidmonocyticleukemiacellsundergoingdifferentiationtomacrophages.Wesys
4、tematicallymeasuretemporalregulationofexpressionandvariationbyprofiling120singlecellsateightdistincttimepoints,andinferhighlycontrolledregulatorymodulesthroughwhichsignalingoperateswithstochasticeffects.Thisrevealsdynamicandspecificrewiringasacellularstrategyfordifferentiation.Theintegrati
5、onofbothpositiveandnegativeco-expressionnetworksfurtheridentifiestheproto-oncogeneMYBasanetworkhingetomodulateboththepro-andanti-differentiationpathways.Conclusions:Comparedtoaveragedcellpopulations,temporalsingle-cellexpressionprofilingprovidesamuchmorepowerfultechniquetoprobeformechanist
