2、ue.,CanadaH3T1E2AbstractApoptosisinmammaliancellscanbeinitiatedthroughtwomajorinterrelatedpathways,oneinvolvingengagementoftheTNFfamilyofdeathreceptors,theotherinvolvingthereleaseofcytochromecfrommitochondria.UnlikeothermembersoftheTNFligandfamily,TNF-relatedapoptosis-inducin
3、gligand(TRAIL)preferentiallyinducesapoptosisintumorcelllines,butnotinnormalcells,suggestingthatTRAILcouldpotentiallyrepresentapowerfulcancertherapeutic.RecentexperimentshaverevealedthatoneofthekeyregulatorsofTRAILexpressioninlymphocytesistheNF-Btranscriptionfactors.SeveralTR
4、AILreceptorshavebeenidentified:twoofthesereceptorsTRAIL-R1/DR4andTRAIL-R2/DR5containcytoplasmicdeathdomainsandsignalapoptosis,whiletwootherdecoyreceptors,TRAIL-R3/DcR1andTRAIL-R4/DcR2lackafunctionaldeathdomainanddonotmediateapoptosis.ManycancercelllinespreferentiallyexpressTRA
