6、OLERO-1,arandomized,double-blindtrialevaluatingtheefficacyandsafetyofaddingeverolimustotrastuzumabandpaclitaxelasfirst-linetherapyforHER2+advancedbreastcancer.Inthistrial,thebenefitofeverolimuswasseenonlyinthepre-definedsubsetofpatientswithhormonereceptor-negativebreastcanceratbaseline(progre
7、ssion-freesurvivalhazardratio=0.66,P=0.0049).Astrictinterpretationofthisfinding,basedoncomplex“alphasplitting”rulestoassessstatisticalsignificance,ledtotheconclusionthatthebenefitofeverolimuswasnotstatisticallysignificanteitheroverallorinthes
