1、—1504—药物分析杂志ChinJPharmAnal2014,34(8)HPLC法测定阿托伐他汀钙有关物质张晓峰,侯晓清,邓凤霞,陈立云,田丽娜(北京嘉林药业股份有限公司,北京100121)摘要目的:建立阿托伐他汀钙有关物质测定方法。方法:采用HPLC法,已知杂质采用对照品法定性和定量,其他杂质采用自身对照法计算。采用WondasilC18色谱柱(4.6mm×250mm,5μm),以乙腈-四氢呋喃-醋酸铵缓冲液为流动相,梯度洗-1脱,流速1.0mL·min,检测波长244nm,柱温30℃,进样量20μL。
2、结果:阿托伐他汀与相邻杂质(杂质B和杂质C)及各已知杂质之间的分离度均大于1.5;杂质A、B、C、D、E的定量限分别为30、40、40、20、10ng,且在各自的线性范围内线性关系良好(r>0.9901,n=5);平均回收率(n=9)分别为98.5%、111.3%、91.5%、106.6%、106.5%,RSD分别为4.8%、4.1%、4.7%、7.3%、6.4%。结论:本法经方法学验证,可测定阿托伐他汀钙中的有关物质。关键词:阿托伐他汀钙;有关物质;杂质检查;去氟阿托伐他汀(杂质A);阿托伐他汀对映体(
