1、CO2对人卵巢癌SKOV3细胞裸鼠皮下移植瘤生长和转移的影响作者:王亚平,张爱荣单位:山东大学第二医院妇产科,济南【摘要】目的探讨不同压力持续性CO2气腹对人卵巢癌SKOV3细胞裸鼠皮下移植瘤生长和转移的影响。方法将体外培养的人卵巢癌细胞株SKOV3分别暴露于不同压力(8、10、12mmHg)和作用时间(1、3h)的CO2环境中,对照组细胞放入CO2培养箱常规培养。收集各组细胞后接种于裸鼠腋下,建立有差异的裸鼠皮下移植瘤模型并绘制肿瘤生长曲线。接种6周后脱臼处死全部裸鼠,测量移植瘤的体积和重量,采用免疫组织化学法检测各组中增殖细胞核抗原(PCN
2、A)和nm23-H1蛋白的表达。结果接种7d后各组均明显成瘤,各CO2气体处理组的皮下移植瘤生长速度均快于对照组,其皮下移植瘤体积和重量也高于对照组(P<0.01)。各CO2处理组中肿瘤细胞PCNA蛋白表达阳性率均高于对照组(P<0.01),各组间nm23-H1蛋白表达阳性率差异无统计学意义(P>0.05)。结论CO2气体本身对人卵巢癌SKOV3细胞裸鼠皮下移植瘤的生长有明显的促进作用,对转移影响不大。【关键词】卵巢肿瘤;二氧化碳;小鼠,裸;移植瘤 Effectofcarbondioxideonthegrowthandmetastasisof
