2、IiY2372192毕业论文题目:亟壁在肠超迦[]菌拉适丝氢氢虫的佳旦佥折生塑丝堂皇坌王生塑堂专业2QQ2级亟±生姓名:萱堂松指导教师(姓名、职称):塞!』查勇(熬握21曾型(麴握2摘要目的:sRNA(smallnon.codingRNA,sRNA)是一种细菌转录的非编码小RNA,在细胞的毒力调控及与宿主的相互关系中起着重要作用。沙门菌可引起人类食物中毒、败血症及伤寒等,从RNA水平研究沙门菌的致病性将为沙门菌的防治提供新方向。方法:本课题以肠炎沙门菌为模型,研究肠炎沙门菌sRNA(istR)在抗活性氮氧中的作用。运用Red同源重组系统对肠炎沙门菌(SE2472)的括绽基因进行敲除
3、,构建敲除菌株(SE2472AistR)。比较野生株和敲除株对活性氮氧的敏感性。结果:实验结果表明,SE2472AistR对酸性亚硝酸钠(pH5.0,20mmol/L)的抑菌作用更为敏感,两者对H202的抑菌作用无明显差异。在此基础上,本试验构建/stR回复质粒,并将其转入istR敲除株,分析istR的回复表达与沙门菌耐活性氮的作用,回复实验结果显示,istR回复表达后,沙门菌抵抗活性氮的作用得以回复。结论:综合上述结果,推测istR在沙门菌抗活性氮中起着重要作用,与抗活性氧作用未有明显联系,对于istR如何调控沙门菌抗活性氮的作用机制还需进一步深入研究。关键词:沙门菌,非编码小R
