1、摘要摘要目的:探讨辛伐他汀对动脉粥样硬化(AS)大鼠主动脉血红素氧合酶1(HO.1)表达的影响。方法:取健康SD大鼠24只,随机分为三组:对照组、AS模型组和辛伐他汀治疗组,每组8只。AS模型组和辛伐他汀治疗组在实验开始时,一次性腹腔注射维生素D360万U/kg,以后喂食高脂饮食;对照组喂食普通饲料。4周后辛伐他汀治疗组灌胃给予辛伐他汀(10mg/kg.d)治疗。第8周末处死大鼠,测定血脂水平,取主动脉观察形态及病理学变化,同时采用免疫组织化学方法检测大鼠主动脉HO一1的表达情况,并应用人工计数分析法对免疫组化结果进行定量分析。结果:(1)AS模型组主动
2、脉出现典型AS斑块,其主动脉HO.1的表达水平显著高于对照组(P(0.。001)。(2)辛伐他汀治疗组的主动脉粥样硬化程度与AS模型组比较明显减轻,但其HO.1的表达水平显著高于AS模型组(P(0.001)。结论:(1)HO.1在AS大鼠主动脉组织中表达上调。(2)辛伐他汀能够上调HO.1在AS大鼠主动脉的表达水平。,(3)辛伐他汀可能通过上调HO.1的表达而发挥抗AS作用。关键词:动脉粥样硬化;大鼠;血红素氧合酶.1;辛伐他汀ⅡAbstractABSTRACTobjective:Tostudytheeffectofsimvastationontheex
