6、······································································34个人简历·············································································35中文摘要Sox2在不同发育阶段SD大鼠穹窿下器细胞的表达摘要目的:观察出生后不同发育阶段大鼠穹窿下器(subfornicalorgan,SFO)的Sox2阳性细胞的形态及分布特征,观察其表达情况。方法:雄性健康SD大鼠,分为幼
7、年组和成年组,每组各24只,清洁级,实验动物不作任何处理。1标本制备常规动物标本制备:将两组实验动物用水合氯醛(0.3ml/100g)麻醉后,自主动脉灌注4%多聚甲醛磷酸盐缓冲液(0.1mol/L,PH7.2)进行前固定,立即取脑,进行后固定。将组织标本经脱水,透明,包埋,选取含穹窿下器部分,石蜡切片机冠状面切片,厚度约为5μm,制成石蜡切片。对实验动物的处置符合动物伦理学要求。2染色常规免疫组织化学染色:将上述石蜡切片经Sox2染色,使用中性树胶封片后,显微镜观察、照相。3统计学处理选取相同SFO的横断面,使用多媒体真彩病理图文
8、分析系统,在20倍镜下,通过计数SFO内细胞总数和Sox2阳性细胞数,计算出阳性细胞率,计量资料均采用x±s表示。结果:1不同发育阶段大鼠Sox2阳性细胞在SFO的分布。幼年组大鼠的SFO,腹侧部的室管膜Sox2阳性细胞排列整齐而密集,中央部的室管
