2、gencymedicine)DissertationSupervisor:Prof.ChengQingHongJune，2015摘要目的：研究HO-1在脓毒症大鼠肺组织中的表达及意义。方法：选取48只SD大鼠随机分成2组，假手术组和脓毒症组。脓毒症组24只，采取盲肠结扎穿孔（CLP）法制造脓毒症模型；假手术组24只，仅做开关腹处理。两组分别于术后6h、12h、24h处死大鼠，迅速开胸留取肺组织，用10％甲醛溶液固定，固定24小时，制备石蜡包块，以备制作HE染色光镜病理切片观察肺组织病理变化。通过Westernblot法测定HO-1蛋白变化，用RealtimePC

3、R法测定HO-1mRNA水平的变化。结果：1.不同检测时间，大鼠的肺组织表现出来的病理特征不同，脓毒症的大鼠比假手术的大鼠的病例变化更加明显，出现了肺内多种问题，例如出血、水肿、炎症等问题；2.不同检测时间点，脓毒症的大鼠体内HO-1的表达水平要明显高于假手术的大鼠；3.手术后的第6h、12h、24h的各个时间点，HO-1mRNA的表达水平在脓毒症组大鼠体内的表达明显高于假手术组，差异具有统计学意义。结论：1.在脓毒症模型组中，大鼠肺组织中的HO-1的表达与对照组存在差异。2.脓毒症组中HO-1的高表达，可提示HO-1参与脓毒症引起的肺损伤。关键词：脓毒症;血红
