6、·············46个人简历·············································································48中文摘要YAP与P-YAP在肝细胞癌中的表达与其临床生物学行为关系的研究摘要目的:肝细胞癌(HCC,hepatocellularcarcinoma)是肝脏原发肿瘤中最常见的类型,也是我国乃至全世界.最常见的恶性肿瘤疾病之一。通常HCC被发现时患者已进入晚期,常规化疗对其作用不明显,致使生存率较其它癌症显著降低。因此,加强对HCC的发病机制研究,寻找更多关于肝癌基因及抑癌基因的表达并明确其与

7、HCC的关系已成为当前研究的重要方向。Hippo通路在促进细胞凋亡,限制细胞增殖,调控器官分化方面起着重要的作用。YAP(Yes-associatedprotein)即Yes相关蛋白,作为该通路下游的效应因子,多以磷酸化形式(P-YAP)参与细胞信号转导以调控细胞凋亡与增殖的平衡。在病理状态下,被阻断或失活的Hippo通路对YAP磷酸化作用失效致YAP自身活性增强使细胞增生和凋亡间平衡失调、组织器官过度生长及肿瘤发生。国内外相关研究表明,YAP作为新近发现的癌基因,参
