6、·44个人简历·············································································45中文摘要MTDH、IκB、NF-κBP65在乳腺癌组织中的表达及意义摘要目的:乳腺癌(breastcancer)已成为当今女性最常见的恶性肿瘤,近年来其发病率和死亡率均呈现上升趋势。乳腺癌好发于45-55岁的女性,然而随着人们生存环境、生活方式以及饮食习惯的改变,发病年龄渐渐趋于年轻化。乳腺癌的形成原因复杂多样,它是一个多因素、多突变积累的结果。关于乳腺癌的发病机制存在很多学说。近几年来

7、,对乳腺癌分子生物学层面的研究已成为此领域研究的热点。MTDH又称为AEG-1、LYRIC,该癌基因位于人类8号染色体上(8q22),在多种肿瘤细胞中高表达,并与肿瘤细胞的增殖、侵袭、转移及耐药密切相关。IκB是核因子κB(NF-κB)的抑制蛋白。NF-κBP65是NF-κB的一个亚单位,它的高表达与肿瘤细胞的侵袭、转移密切相关。在细胞受到刺激时,三者通过共同作用诱导相关基因转录,促进肿瘤的发生发展。本实验通过实时荧光定量PCR及免疫组织化学法分别检测MTDH、IκB、NF-κBP65三者在乳腺癌组织及癌旁正常组织(距离癌灶>5cm)中mRNA及蛋白质含量
