1、摘要油菜是我国主要的油料作物,大约有50%的食用植物油来源于油菜。长期以来,选育高产优质油菜一直是育种家所面临的主要任务。实践证明,杂种优势的利用是提高油菜产量的主要途径,而雄性不育是杂种优势利用的重要手段。隐性细胞核雄性不育因其具有不育性彻底、稳定、恢复源广等优点,在国内已经成为杂种优势利用的一个重要途径,但由于得不到100%的不育系,在杂交制种时需人工拔除不育系株行中50%的可育株,限制了其广泛应用。本研究以甘蓝型油菜双隐性细胞核雄性不育系S45AB为材料,利用抑制性差减杂交技术(SSH)构建了S45AB
2、小花蕾期的正反向差减文库。对差减文库进行质量检测表明其质量完好。藉此,可以通过反向Northern杂交技术筛选到育性相关基因,分析S45AB雄性不育机理,为更好地利用S45AB打下坚实的基础。另外,我们利用双链特异性核酸酶(duplex.specificnuclease,DSN)均一化方法构建了甘蓝型油菜华杂4号小花蕾的均一化文库。对均一化文库进行评价结果表明其均一化效果理想。利用这个文库,可以制备油菜基因芯片,来进行油菜功能基因组研究。关键词:甘蓝型油菜;隐性核不育;抑制性消减杂交;均一化cDNA文库;双链

6、ardlibraries,whichwerehighlyenrichedformalefertility.relatedgenefragmentsofrecessivegeniemalesterility(RGMS)two‘typelinesS45ABlittlebuds,wereconstructedusingsuppressionsubtractivehybridization(SSH).QualitycheckshowedthatqualityofsubtractivelibrariesWasgood.
