1、多发性骨髓瘤患者外周血单个核细胞c-Cbl和Cbl-b基因表达下降方苏1,李萡1*,牛宇哲1,2,陈少华1,杨力建1,李扬秋1,2(510632广州,暨南大学:血液病研究所1;再生医学教育部重点实验室2)[摘要]目的了解多发性骨髓瘤(multiplemyeloma,MM)患者外周血单个核细胞中c-Cbl和Cbl-b基因表达水平。方法利用SYBRGreen实时定量PCR方法,检测16例MM患者和12例健康人外周血单个核细胞c-Cbl和Cbl-b基因的表达情况,以β2微球蛋白基因(β2M)作为内参照,采用2-ΔCt×100%计算c-Cbl和Cbl-b基因相对表达水
2、平。结果MM患者中c-Cbl和Cbl-b基因表达水平与健康人相比均下降,其中MM组c-Cbl(平均秩:12.56)表达水平与健康对照组(平均秩:17.08)比较无统计学意义(P=0.159);而Cbl-b(平均秩:10.72)表达水平与健康对照组(平均秩:19.54)则显著降低(P<0.05﹚。结论c-Cbl和Cbl-b表达下调可能通过影响BCR和TCR信号传导通路从而影响机体的免疫调节功能。[关键词]多发性骨髓瘤(MM);c-Cbl;Cbl-b;荧光定量PCR[中图法分类号][文献标志码]ADecreasedc-CblandCbl-bgenesexpress
