1、提要目的:通过观察大黄及大黄氯仿提取物对实热证大鼠一般状况、寒热表征、能量代谢的影响,分析其作用趋势、特点及规律,探寻寒性中药对热性疾病机体能量代谢的作用规律及机制,为更好的指导临床用药提供数据参考。方法:选Wistar大鼠,皮下注射2,4-二硝基苯酚造大鼠实热证模型,观察模型大鼠给予大黄水煎液、大黄氯仿提取物溶液后的一般状况变化、自主活动、寒热趋向状况;采用紫外分光光度法检测肝糖原、丙酮酸、甘油三酯、ATP酶活力、琥珀酸脱氢酶活力、乳酸脱氢酶活力;酶联免疫吸附法检测大鼠血浆T3、T4、TSH、TRH水平;采用高效液相色谱法检测肝组织ATP含量并计算肝细胞能荷
2、值。结果:与模型对照组比较,大黄水煎液能显著降低实热大鼠体温,降低血浆中乳酸、丙酮酸、甘油三酯含量和甲状腺激素水平;降低肝组织糖原、乳酸、丙酮酸含量;抑制琥珀酸脱氢酶(SDH)、乳酸脱氢酶的活性(LDH)及ATP酶活性;降低能荷值。大黄氯仿提取物能显著降低实热大鼠体温,降低血浆中乳酸含量和T4水平;降低肝组织糖原、丙酮酸含量;抑制琥珀酸脱氢酶(SDH)活性;降低肝组织能荷值。结论:大黄水煎液、大黄氯仿提取物对实热证大鼠机体的能量代谢有一定的抑制作用,这为今后大黄治疗热性疾病的研究和临床应用提供了有价值的数据参考。关键词大黄;大黄氯仿提取物;大鼠实热证模型;能量
