1、#临床药学#$HPLC法测定肾移植患者血浆中霉酚酸的浓度叶丽卡,谢志红,王若伦,许欣,张伟婷(广州医学院第二附属医院,广州市510260)中图分类号R96911文献标识码A文章编号1001-0408(2008)17-1306-03摘要目的:建立以高效液相色谱法测定霉酚酸血药浓度的方法,并研究其在中国肾移植患者体内的多剂量药动学特征。方法:血-1-1浆采用乙腈蛋白沉淀后进样。其中,色谱柱为DiamonsilC18,流动相为乙腈-10mmol#LKH2PO4(5B6),流速为111mL#min,检测波长为254nm,柱温为40e。此方法应用于测定12例肾移植患者的多剂量药动学。结
2、果:色谱中霉酚酸与内标分离良好,血浆-1-1中的内源性杂质无干扰。霉酚酸检测浓度在0138~59100Lg#mL范围内线性关系良好,最低检测限为0138Lg#mL;回收率为89132%~97163%,日内、日间RSD均<8100%。药动学结果显示患者服用霉酚酸酯后,霉酚酸的药动学个体差异较大,与文献报道基本一致。结论:本方法结果准确,操作简便,适合于霉酚酸的药动学研究。关键词霉酚酸;高效液相色谱法;肾移植患者;血药浓度;多剂量;药动学DeterminationofMycophenolicAcidinKidneyTransplantationPatientsbyHPLCYELi
