1、湖北中医药大学硕士学位论文“瓜蒌--薤白”药对对大鼠心肌缺血再灌注损伤保护作用的研究姓名:张承志申请学位级别:硕士专业:药理学指导教师:但汉雄20120528中文摘要目的:采用《金匮要略》中张仲景治疗胸痹的基础方一瓜蒌薤白白酒汤,通过古法制备,观察瓜蒌薤白配伍前后对大鼠缺血再灌注损伤的保护作用,探讨“瓜蒌一薤白∞药对抗心肌缺血再灌注损伤的作用机制,从而为进一步开发“瓜蒌一薤白"药对的药用价值,为临床安全、有效、合理使用“瓜蒌一薤白"药对,治疗胸痹(冠心病)提供科学、客观的实验依据。方法:在体结扎大鼠冠状动脉左前降支法,造成心肌缺血,
2、30min后松开活结,恢复血流灌注60min,造成心肌缺血再灌注模型,设瓜蒌薤白复方组和单味药组,并设有阴性对照组和阳性对照组;(1)以心电图标准导联II的ST段电位值的变化为指标,观察各组对大鼠心肌缺血再灌注心电图的改善作用;(2)测定心肌缺血再灌注大鼠的血清肌酸激酶同工酶(CK-MB)和乳酸脱氢酶(LDH)水平,以及血清丙二醛(MDA)、超氧化物歧化酶(SOD)的水平;(3)采用伊文思蓝和TTC双染色法确定缺血区和梗死区面积;(4)对心肌组织进行HE染色观察心肌坏死组织的病理变化和用电镜观察大鼠心肌组织超微结构的变化;(5)We
