1、重庆医科大学硕士学位论文大黄素下调ERCC1和Rad51对非小细胞肺癌增殖的影响及分子机制姓名:何林申请学位级别:硕士专业:病理学与病理生理学指导教师:叶秀峰201205重庆医科大学硕士研究生学位论文大黄素下调ERCCl和Rad51对非小细胞肺癌增殖的影响及分子机制摘要目的:研究大黄素对非小细胞肺癌的细胞毒性,并观察其对非小细胞肺癌ERCCl和Rad51表达的影响,探讨大黄素对非小细胞肺癌增殖的影响及其机制。方法:体外培养肺腺癌细胞株A549和肺鳞癌细胞株SK.MES一1,经大黄素按浓度梯度:O、
2、25、50、75、100岬ol/L处理48h。MTT法测定大黄素对A549和SK.MES.1细胞的增殖抑制作用。用RT.PCR和Westemblot法测定目的基因RECCl和Rad51的表达情况。透射电镜观察大黄素处理A549和SK.MES.1细胞前后的超微结构变化。结果:MTT法测定出大黄素作用48h时A549和SK.MES.1细胞株IC50值分别为(70.18±1.935)岬ol/L和(41.95±1.266)岬ol/L。大黄素对A549和SK.MES.1细胞的增殖抑制作用呈剂量依赖性。RT—
