2、lyenlargemyocardialinfarctionsize,increasethemortalityandaffectetheprognosisofthepatientswithAMI.ApoptosisWasbelievedtobeanimportantwaythatcardiacdiedafterAMI,anditmarkedlyaffectedmyocardialinfarctionsize,theventricularfunctionandtheprognosisofthepatientswithAMI.Thepresent

3、studyaimedtoobservetheeffectofacutehyperglycaemiaonapoptosisanddiscusseditspossiblemechanismbyreproducingtheanimalmodelofAMlwithhyperglycaemia.MethodsThemodelofAMlwasreproducedbyligatingtheleftanteriordescendingcoronaryartery.Then,theforty-sevenmaleSDratswererandomlydivide

4、dinto4groups:controlgroup:theratsweretreatedbyintravenousinfusionofphysiologicalsalineafterAMI;HGlgroup:theratsreceivedintravenousinfusionof25%glucosesolutionafterAMI;HG2group:theratsweretreatedbyintravenousinfusionof50%glucosesolutionafterAMI;Insulingroup:theratsreceivedi

5、ntravenousinfusionof50%glucosesolutionafterAMIand5unitregularInsulinWassubcutaneoulyinjectedatonce.AnothertenmaleSDratsweresupposedasshamgroup:theoperationissimilarexceptforligatingtheleftanteriordescendingcoronaryarteryandthephysiologicalsalinewasintravenouslyinfusedafter

6、theoperation.Thefluidwasinfusedbyapumpatthespeedof4ml/kg’h,continuouslyfor30minineverygroup.BloodglucoselevelsWL。l-emonitoredthroughouttheexperiment.Attheendoftheexperiment,themyocardialsampleswastakentomeasuretheindicators.Thehistopathologicalchangesofhearttissueswasobese

7、rvedwithHEstainingbylight·-microscopy;theexpressionofcaspase·9wasmeasuredbyimmunohistochemistry;theapoptoticindex(AI)andMitochondriaMembrancePotentialwasdeterminedbycorrespondingkitseparately.TheexpressionofCyt-Cinmitochondriaandendochylemaweredeterminedbywestemblotting.5◆
