1、中文摘要人工合成E—选择素对大鼠局灶性脑缺血再灌注损伤脑组织血管细胞粘附分子一1及其mRNA表达的影响第二部分:人工合成E一选择素对大鼠局灶性脑缺血再灌注损伤后血管细胞粘附分子一1蛋白和mRNA表达的影晌目的:探讨人工合成E进择素在对大鼠局部脑缺血再灌注损伤中脑组织血管细胞粘附分子一1及其mRNA表达的影响及其作用机制。方法:42只雄性SD大鼠被随机分为假手术组、模型组和人工合成E进择素治疗组。采用改良的ZeaLonga法建立脑缺血再灌注损伤模型,治疗组大鼠用人工合成E-;iz'垂择素10mg/Kg股静脉注射体内。免疫组化法观察缺血区脑组织中微血管
2、的血管细胞粘附分子一1(vascularcelladhesionmolecule一1,VCAM一1)阳性细胞数,RT-PCR法观察其mRNA表达。结果:1.假手术组:VCAM一1阳性细胞数少,VCAM一1mRNA表达低。2.模型组:VCAM一1阳性细胞在再灌注2h后开始出现,并持续增多,在24h后达高峰,72h开始下降,各时间点与假手术组比较明显增多(P<0.01);VCAM—ImRNA在再灌注2h后开始表达,12h后达到高峰,24h开始降低,各时间点与假手术组比较明显增高(P<0.01)。3.治疗组:VCAM一1阳性细胞数较假手术组多(P<0.0
