2、对照组),采用流式细胞术检测外周血及脾脏CD4+CD25+CDl2710w/-Treg的表达情况。将新鲜的脾脏组织冲洗干净,取一小块组织(约10mm3),用Medimachine系统制备单细胞混悬液,并用PBS稀释至0.5万个/此。每例标本取两个试管,分别加入CD4-FITC/mouseIgGlPE,CD4.FITC/CD25.PE/CDl27.AlexaFluor@647各10I.tL:每管再加入PBS稀释后的混悬液601xL后混匀避光孵育25min;jJl:l2mLl×LysingBuffer,混匀避光孵育15min;溶血后PBS洗2次,最后加200斗
