1、青岛大学硕士学位论文姜黄素对前列腺癌PC3细胞和肺癌H446细胞Id1表达的影响姓名:徐菲菲申请学位级别:硕士专业:病理学与病理生理学指导教师:于小玲20120529姜黄素对前列腺癌PC3细胞和肺癌H446细胞ldlmRNA表达的影响摘要背景随着对肿瘤分子机制研究的深入,近年来Idl(inhibitorofdifferentiationorinhibitorofDNAbinding,细胞分化抑制因子i)蛋白与人类恶性肿瘤的关系已成为研究的热点;Idl蛋白的过度表达与肿瘤的发生、发展密切相关。体外研究表明,在多种肿瘤细胞系中Idl呈高表达,国内外研
2、究均证实Idl在体内多种恶性肿瘤中有过度表达,包括人的前列腺癌和肺癌组织中Idl也呈高表达。姜黄素是近年来治疗肿瘤研究的热点中药之一,姜黄素是否影响前列腺癌和肺癌细胞中Idl的表达目前国内外均未见报道。目的从细胞水平观察探讨姜黄素对雄激素非依赖性前列腺癌PC3细胞和小细胞肺癌H446细胞中Idl基因表达的影响,以及对PC3细胞和H446细胞的增殖、迁移和凋亡的影响,为临床前列腺癌和肺癌的有效治疗提供相应的理论依据。方法用实时荧光RT-PCR法检测姜黄素对PC3和H446细胞中IdlmRNA表达的影响。分别采用MTT法检测PC3和H446细胞生长活
